- Compatibility Study of Ambroxol HCl Drug-Excipients by Using IR Spectroscopy

Compatibility must be established between the active ingredient and other excipients to produce a stable efficacious, attractive and safe product.
In this report FTIR studies were performed and major peaks were determined by the (FTIR-8400S Shimadzu, Japan) will show band shift and broadening compared to the spectra of pure drug. To study the interaction between AmbroxolHCl, selected excipients (sorbitol, mannitol, sucrose).IR spectra of Ambroxol hydrochloride bands were found at 1595-1545 cm-1 for aromatic C=C stretching, 3350-3250cm-1 for aromatic primary amine NH stretch, 3400-3345 cm-1 for intermolecular hydrogen bonded OH stretch and 700-600 cm-1 for C-Br stretching. This confirms the identity of Ambroxol hydrochloride.

The studies prior to the formulation are broadly known as preformulation studies and these studies should focus on those physicochemical properties of compounds that could affect drug performance and development of an efficacious and specific dosage form. The preformulation studies were carried out of Ambroxol HCl in terms of tests for identification (physical appearance, melting point and IR spectra), solubility profile and quantitative estimation of drug by considering formulation of medicated chewing gum (MCG).Keywords

Out of which Medicated chewing gum delivery system is convenient, easy to administer - anywhere, anytime - and is pleasantly taste making it patient acceptable. It offers a wide range of advantages that make it an excellent alternative. The present work deals with optimization of designed MCG , so that the best possible set of parameters affecting result can be selected to get the desired output. In this report nine batches of formulations (F1-F9) prepared and studied. Two variables were selected for the formulations in which first was concentration of elastomer and other was concentration of softener. The mathematical models developed for all the dependent variables using statistical analysis software Design experts 8.0.1.Percent Drug release of various formulation F2, F5, F8 andF9 was found to be 95.31, 98.74, 88.18 and 91.74. The highest Percent of Drug release among all is of F5.Keywords
